South China Agricultural University developed drug-loaded contact lenses for fungal keratitis

Jiang Gangyu, a professor at South China Agricultural University, designed and prepared a drug-loaded contact lens capable of long-acting sustained-release voriconazole, which achieved local high-efficiency administration of cornea with low toxicity side effects on fungal keratitis. Related research has recently been published online in the journal American Chemical Society Nano.

Fungal keratitis is a serious disease. If the diagnosis and treatment are not timely, it will lead to corneal perforation, blindness, and more serious eyeball removal. Among the existing drugs, voriconazole has a significant effect on the treatment of fungal infections, but it is difficult to dissolve in water. Clinically, there are no voriconazole eye drops or ointment-type products, and only oral tablets and systemic injection preparations have large side effects.

The research group used natural polymer chitosan quaternary ammonium salt derivatives as film-forming materials, and enhanced the mechanical properties and flexibility through cross-linking of carboxylated graphene, and blended with nano-silver to prepare contact lenses with good transparency. Graphene is easy to adhere to the advantages of benzene ring drugs, and the poorly soluble voriconazole is supported on carboxylated graphene, synergistically enhancing the inhibitory ability of voriconazole against fungal attack and the broad-spectrum antibacterial function of quaternary ammonium salt and nano silver. A drug nano-release delivery system that has specific effects on fungal keratitis has been obtained, which solves this general clinical problem.
